All the winners’ goals came from foreigners with Nigerian Morice Chukwu and Kenyan Duke Abuya also netting for the home side.
Singida led 3-0 at half-time and Abuya added a fourth goal a minute after half-time before Abdallah Saleh notched a consolation goal.
Both Tchakei goals and the one from Saleh came off penalties in the preliminary round first leg encounter.
The match was staged in the Tanzanian commercial capital, Dar es Salaam, and the return match on August 27 will also be played there with JKU ceding home advantage for an undisclosed reason.

In the round of 32, Singida or JKU will face Future, a rising force in Egypt who reached the Confederation Cup group stage last season.
A further 10 first legs are scheduled for Saturday and six for Sunday in the African equivalent of the UEFA Europa League.
Among the favourites to lift the trophy are title-holders USM Alger of Algeria and former winners FUS Rabat and Renaissance Berkane of Morocco and Zamalek of Egypt.
